Double stains of living actually translocating sieve elements.double stains

The direction of fluorochrome translocation is from right to left in (A) and (B).

(A) Double stain with locally applied RH-160 (red) and translocated CDCFDA (green). CDCFDA remains limited to the sieve elements and companion cells. The sieve plate seem to be unplugged.

(B) Double stain with locally applied RH-160 (red) and translocated HPTSA (green). The parietal protein deposition (arrows) against the sieve plate does not impede the translocation of HPTSA. A crystalline P protein body (asterisk) is stained by HPTSA. P plastids (arrowheads) are parietally and evenly positioned.
